ANALYSIS OF SPERM KINETICS, PLASMATIC MEMBRANE INTEGRITY AND RESISTENCE TO OXIDATIVE STRESS IN EQUINE FROZEN SEMEN WITH DIFFERENT SPERM CONCENTRATIONS

The cryopreservation process of equine semen involves many proceedings which includes the arrangement of sperm cells in a determinate concentration. However in the literature is related the use of values comprised in a large range of concentration per volume unity. Hence the present experiment evaluated the effect from different sperm concentrations about some variables of seminal quality after thaw. Four ejaculates of five stallions were collected, from breed various (n=20) and each ejaculate was frozen in straws of 0,5mL in concentrations of 100, 200 and 400x106 sptz/mL. It were evaluated the characteristics of sperm movement over the computer analysis (CASA), plasmatic membrane integrity that employs the fluorescent probes of propidium-iodide and carboxyfluorescein diacetate and susceptibility of lipid peroxidation by the technique from quantification thiobarbituric acid reactive substances (TBARS). It was observed the significant effect of concentration (p0,05). Therefore by token of the variables from sperm kinetics it was observed that the freezing of equine semen using lower concentrations should become a benefic practice, however it should be made fertility tests to evaluate the real superiority of this proceeding and the biologic meaning, because when others variables like plasmatic membrane integrity and susceptibility of oxidative stress were evaluated, it were not observed effects that justify the adoption of this proceeding.
